RESUMO

Pneumonia can seriously threaten the life of forest musk deer (FMD, an endangered species). To gain a comprehensive understanding of pneumonia pathogenesis in FMD, iTRAQ-based proteomics analysis was performed in diseased (Pne group) lung tissues of FMD that died of pneumonia and normal lung tissues (Ctrl group) of FMD that died from fighting against each other. Results showed that 355 proteins were differentially expressed (fold change ≥ 1.2 and adjusted P-value < 0.05) in Pne vs. Ctrl. GO/KEGG annotation and enrichment analyses showed that dysregulated proteins might play vital roles in bacterial infection and immunity. Given the close association between bacterial infection and pneumonia, 32 dysregulated proteins related to Staphylococcus aureus infection, bacterial invasion of epithelial cells, and pathogenic Escherichia coli infection were screened out. Among these 32 proteins, 13 proteins were mapped to the bovine genome. Given the close phylogenetic relationships of FMD and bovine, the protein-protein interaction networks of the above-mentioned 13 proteins were constructed by the String database. Based on the node degree analysis, 5 potential key proteins related to pneumonia-related bacterial infection in FMD were filtered out. Moreover, 85 dysregulated proteins related to the immune system process were identified given the tight connection between immune dysregulation and pneumonia pathogenesis. Additionally, 12 proteins that might function as crucial players in pneumonia-related immune response in FMD were screened out using the same experimental strategies described above. In conclusion, some vital proteins, biological processes, and pathways in pneumonia development were identified in FMD.

BACKGROUND: Forest musk deer is an endangered species globally. The death of captive forest musk deer can be caused by certain respiratory system diseases. Acute respiratory distress syndrome (ARDS) is a huge threat to the life of forest muck deer that breed in our department. METHODS: Lung histopathologic analysis was conducted by hematoxylin and eosin (HE) staining. The lung gene changes triggered by ARDS were examined by RNA sequencing and related bioinformatics analysis in forest musk deer. The potential functions of unigenes were investigated by NR, SwissProt KOG, GO, and KEGG annotation analyses. Vital biological processes or pathways in ARDS were examined by GO and KEGG enrichment analyses. RESULTS: A total of 3265 unigenes were differentially expressed (|log2fold-change|> 2 and adjusted P value < 0.01) in lung tissues of 3 forest musk deer with ARDS compared with normal lung tissues of the non-ARDS group. These differentially expressed unigenes (DEGs) played crucial roles in immunity and defense responses to pathogens. Moreover, we identified the DEGs related to one or more of the following biological processes: lung development, immunity, and bacterial/viral/fungal infection. And six DEGs that might be involved in lung injury caused by immune dysregulation or viral/fungal infection were identified. CONCLUSION: ARDS-mediated lung gene alterations were identified in forest musk deer. Moreover, multiple genes involved in lung development and lung defense responses to bacteria/viruses/fungi in ARDS were filtered out in forest musk deer.

Improving the photocatalytic performance of metal-organic frameworks (MOFs) is an important way to expand its potential applications. In this work, zero-dimensional (0D) Bi2O3 nanoparticles were anchored to the surface of tridimensional (3D) MIL101(Fe) by a facile solvothermal method to obtain a novel 0D/3D heterojunction Bi2O3/MIL101(Fe) (BOM). The morphology and optical properties of the as-prepared Bi2O3/MIL101(Fe) composite were characterized. The photocatalytic activity of the synthesized samples was evaluated by degrading chlortetracycline (CTC) under visible-light irradiation. The obtained BOM-20 composite (20 wt % Bi2O3/MIL101(Fe)) exhibits the highest photocatalytic activity with CTC degradation efficiency of 88.2% within 120 min. The degradation rate constant of BOM-20 toward CTC is 0.01348 min-1, which is 5.9 and 4.3 times higher than that of pristine Bi2O3 and MIL101(Fe), respectively. The enhanced photocatalytic activity is attributed to the formation of a Z-scheme heterojunction between Bi2O3 and MIL101(Fe), which is conducive to the rapid separation of photogenerated carriers and the enhancement of photogenerated electron and hole redox capacity. The intermediate products were analyzed by liquid chromatography-mass spectrometry (LC-MS), and a possible photocatalytic degradation path of CTC was proposed. This work provides a new perspective for the preparation of efficient MOF-based photocatalysts.

Thermophoresis of charged spheroids has been widely applied in biology and medical science. In this work, we report an analysis of the anisotropic thermophoresis of diluted spheroidal colloids in aqueous media for extremely thin EDL cases. Under the boundary layer approximation, we formulate the thermophoretic velocity, the thermophoretic force, and the thermodiffusion coefficient of a randomly dispersed spheroid. The parametric studies show that under the aforementioned conditions, the thermophoresis is anisotropic and its thermodiffusion coefficient should be considered as a vector, DT . The thermodiffusion coefficient values and directions of DT are strongly related to the aspect ratio and the angle θ between the externally applied temperature gradient and the particle's axis of revolution: The increasing aspect ratio enlarges the thermodiffusion coefficient value DT of prolate (oblate) spheroids to a constant value when θ < 60° (θ > 45°), and it reduces DT of prolate (oblate) spheroids to a constant value when θ > 60° (θ < 45°). The thermodiffusion coefficient direction of both prolate and oblate spheroids deviates slightly from -∇T∞ for a small aspect ratio, and such deviation becomes serious for a large aspect ratio.

Alpine musk deer, Moschus chrysogaster, a solitary, primitive ungulate inhabiting high elevation areas (3000-4500 m) is an endangered species facing threat of extinction globally due to excessive hunting for its musk. In this study, we determined the complete mitochondrial genome of M. chrysogaster, which was 16,354 bp in length, and revealed the same gene order and genomic organization as typical Moschidae mitochondrial DNA. Start codons in 13 protein-coding genes (PCGs) were all typical ATGs except ATA for ND2 and ND3 and ATT for ND5. Stop codons were all typical types except an incomplete stop codon T for COX3, ND2, ND3, and ND4. Secondary structures in 22 transfer RNA genes all showed typical cloverleaf except tRNA-Ser (AGY), in which the dihydrouridine arm formed a simple loop. No repeat units were found in the control region. The topology structure indicated that M. cupreus was primitive and located at the root of the Moschidae clade. Phylogenetic reconstruction placed M. chrysogaster as a distinct lineage, closely related to the branch of M. leucogaster, M. berezovskii (wild) and predicted a sister relationship with M. moschiferus, M. anhuiensis, and M. berezovskii (captive). However, we suggested that the genetic resources of M. chrysogaster_JQ608470 should be further investigated.

Both natural conditions and anthropogenic factors affect the survivability, distribution, and population density of wildlife. To understand the extent and how these factors drive species distributions, a detailed description of animal movement patterns in natural habitats is needed. In this study, we used satellite telemetry to monitor elevational ranges favored by endangered golden snub-nosed monkeys (Rhinopithecus roxellana), in the Qinling Mountains, central China. We investigated the abundance and distribution of food resources through sampling vegetation quadrats at different elevations and sampled anthropogenic activities using field surveys. Our results indicated that although there was no significant variation in food resources between low- (<1500 m) and middle-elevations (1500-2200 m), monkeys were found most often in areas above 1500 m, where there was less anthropogenic development (e.g. houses and roads); however, monkeys rarely ranged above 2200 m and had limited food availability at this altitude. There was limited human disturbance at this elevation. We suggest that both human activity and ecological constraints (i.e. food resources) have considerable effects on elevational use of R. roxellana in the Qinling Mountains. This study highlights the critical roles these factors can play in shaping the vertical distribution of high-altitude primates. This research provides useful insights for habitat-based conservation plans in which human disturbance management and habitat restoration should be prioritized.

Melatonin reportedly increases chilling tolerance in postharvest peach fruit during cold storage, but information on its effects on cell-wall disassembly in chilling-injured peaches is limited. In this study, we investigated the role of cell-wall depolymerization in chilling-tolerance induction in melatonin-treated peaches. Treatment with 100 µM melatonin alleviated chilling symptoms (mealiness) characterized by a decrease in fruit firmness and increase in juice extractability in treated peaches during storage. The loss of neutral sugars, such as arabinose and galactose, in both the 1,2-cyclohexylenedinitrilotetraacetic acid (CDTA)- and Na2CO3-soluble fractions was observed at 7 days in treated peaches, but the contents increased after 28 days of storage. Atomic-force-microscopy (AFM) analysis revealed that the polysaccharide widths in the CDTA- and Na2CO3-soluble fractions in the treated fruit were mainly distributed in a shorter range, as compared with those in the control fruit. In addition, the expression profiles of a series of cell-wall-related genes showed that melatonin treatment maintained the balance between transcripts of PpPME and PpPG, which accompany the up-regulation of several other genes involved in cell-wall disassembly. Taken together, our results suggested that the reduced mealiness by melatonin was probably associated with its positive regulation of numerous cell-wall-modifying enzymes and proteins; thus, the depolymerization of the cell-wall polysaccharides in the peaches treated with melatonin was maintained, and the treated fruit could soften gradually during cold storage.

Moschus berezovskii is an endangered species, but its captive populations are valuable on musk secretions in traditional Chinese medicine and perfume manufacture. The mitogenome of M. berezovskii was 16,353 bp in size. Stop codons in 13 PCGs were all typical types except incomplete stop codon T for COX3, ND2 and ND4, and TA for ND3. No tandem repeat was found in control region. Phylogenetic analysis indicated that Moschidae has the closest relationship with Bovidae. We supported that M. berezovskii should be categorized into two subspecies, and suggested that the status of M. chrysogaster JQ608470 should be further investigated.

Golden snub-nosed monkey (Rhinopithecus roxellana) is one of the most endangered primate species found in China, exhibiting multilevel society consisting of several one-male-females together with their offspring units (OMU), and all-male units (AMU). Female dispersal patterns of the species within herd have been well documented, whereas those of the males within or between herds are still poorly understood. Our results based a long-term observation indicate that more than half of sub-adult males, and half of the deposed males that stayed a short period in OMU disperse between herds, three of them established their own OMU in new herd after the dispersal. Smaller number of the sub-adult and adult males, compared with adult females, stayed in natal herd, implying sub-adult males started dispersing and male-biased dispersal occurred between herds. High frequencies of resident males were wounded as their OUMs were taken over, and resident males co-operation defend bachelor males were found. Mating competition among males within the herd may have contributed to the scenarios of male-biased dispersal. The results also suggest that maintaining connection between isolated herds and establishing the corridors among the fragmented habitats for the species will greatly benefit increasing its gene flow and promoting conservation status.

To understand better the regulatory mechanism of the carotenoid accumulation, the expression profile of relevant carotenoid genes and metabolites were compared between two peach cultivars with different colors during fruit development. Meanwhile, the change pattern of carotenoid content and expression of carotenoid metabolic genes in peaches after harvest in response to blue light were also investigated. As compared to the yellow fleshed-cultivar 'Jinli', lower carotenoid levels were observed in skin and pulp in white peach cultivar 'Hujing', which might be explained by differentially expression of PpCCD4 gene. With respect to 'Jinli', the carotenoid accumulation during fruit development in fruit skin was partially linked with the transcriptional regulation of PpFPPS, PpGGPS, PpLCYB and PpCHYB. However, in the pulp, the accumulation might be also associated with the increased transcriptions of PpPDS, along with the above four genes. Blue light treatment induced carotenoid accumulation in 'Jinli' peaches during storage. In addition, the treated-fruit displayed higher expression of all the eight genes analysed with a lesser extent on PpCCD4, which suggested that the much more increased carotenoid synthesis rate could result in the higher carotenoid content in blue light-treated fruit. The results presented herein contribute to further elucidating the regulatory mechanism of carotenoid accumulation in peach fruit.

The effect of exogenous melatonin on chilling injury in peach fruit after harvest was investigated. To explore the optimum concentration of melatonin for chilling tolerance induction, peach fruit were treated with 50, 100, or 200 µM melatonin for 120 min and then stored for 28 days at 4 °C. The results showed that application of melatonin at 100 µM was most effective in reducing chilling injury of peach fruit after harvest. Peaches treated with melatonin at this concentration displayed higher levels of extractable juice rate and total soluble solids than the non-treated peaches. In addition, melatonin treatment enhanced expression of PpADC, PpODC, and PpGAD and consequently increased polyamines and γ-aminobutyric acid (GABA) contents. Meanwhile, the upregulated transcripts of PpADC and PpODC and inhibited PpPDH expression resulted in the higher proline content in melatonin-treated fruit compared to the control fruit. Our results revealed that melatonin treatment may be a useful technique to alleviate chilling injury in cold-stored peach fruit. The chilling tolerance of harvested peaches induced by melatonin treatment is associated with higher levels of polyamine, GABA, and proline. These data provided here are the first protective evidence of exogenous melatonin in harvested horticultural products in response to direct chilling stress.
